Ejaculate Characteristics of Rabbits Infected with Trypanosoma congolense and Changes Caused after Treatment with Diminazene aceturate (Diminaveto®) International Journal of Morphology
Leigh,Olufisayo Oluwadamilare; Fayemi,Oluremi Eric.
The effects of experimental Trypanosoma congolense infection on the ejaculate of rabbits and changes caused after treatment with Diminaveto® were investigated using 24 New Zealand White rabbits (bucks). The bucks were housed singly in standard rabbit cages and fed on specialized ration containing 10% Protein supplement, grains, legume, salt and fresh water ad libitum during the study. Data on ejaculate characteristics were collected from all the bucks in the first phase (i.e. before infection) and in the second phase (i.e. during infection, with 4.8x10(5) Trypanosoma congolense, intraperitoneally). Similar data were collected from 12 randomly selected bucks treated with 7.0mg/kg Diminaveto® following reconstitution during the third phase. Data collected...

Establishment of an in vitro Transgene Expression System in Epimastigotes of Trypanosoma congolense OAK
Sakurai, Tatsuya; Tanaka, Miho; Shin-ichiro, Kawazu; Inoue, Noboru; 井上, 昇.
The epimastigote form (EMF) of Trypanosoma congolense appears in the late tsetse infective stage. Epimastigotes adhere to the tsetse proboscis, proliferate in this region, and differentiate into mammal-infective metacyclic forms (MCFs). This differentiation is called metacyclogenesis and is indispensable for cyclical transmission of the parasite. Although an in vitro culture method reproducing metacyclogenesis was established several decades ago, few genetic tools have been utilized to elucidate the molecular mechanisms underlying T. congolense metacyclogenesis. In this study, we established a transgene expression system in the EMF of T. congolense IL3000; the EMF was successfully cultured and observed to undergo metacyclogenesis in vitro. The newly...

Establishment of ATP-Based Luciferase Viability Assay in 96-Well Plate for Trypanosoma congolense OAK
SUGANUMA, Keisuke; ALLAMANDA, Puttik; HAKIMI, Hassan; ZHOU, Mo; ANGELES, Jose Ma.; KAWAZU, Shin-ichiro; INOUE, Noboru; 河津, 信一郎; 井上, 昇.
Animal African trypanosomosis (AAT), caused by Trypanosoma congolense, is widespread throughout sub-Saharan Africa. There are significant concerns related to the current drugs available for the treatment of AAT due to their limited effectiveness across species and their adverse effects. Moreover, drug resistant trypanosomes have recently been reported in the field. High throughput screening (HTS) of large chemical compound library collections is a promising approach for identifying novel drug candidates. While HTS for Trypanozoon trypanosomes, T. brucei sspp. and T. evansi is well established, no assays have been developed for T. congolense. In the present study, the authors developed an ATP-based luciferase viability assay for T. congolense in a 96-well...

Expression of a Gene Encoding Trypanosoma congolense Putative Abc1 Family Protein is Developmentally Regulated(Parasitology) OAK
BATICADOS, Waren N.; WITOLA, William H.; INOUE, Noboru; KIM, Jung-Yeon; KUBOKI, Noritaka; XUAN, Xuenan; YOKOYAMA, Naoaki; SUGIMOTO, Chihiro.
During the attempt to seek T. congolense species-specific diagnostic antigens, we discovered one cDNA clone (P74) encoding 74 kDa putative abc1 protein (p74) from T. congolense PCF cDNA library. It has been suggested that members of the abc1 family are novel chaperonins and essential for both the mitochondrial electron transfer in the bc 1 complex and the coenzyme Q biosynthesis. Although abc1 protein in yeast has a nuclear or mitochondrial subcellular location, neither nuclear localization signal nor mitochondrial targeting signal was found within p74. Northern blot analysis revealed that the transcription level of P74 mRNA in bloodstream form (BSF) cells were 4 times higher than that in procyclic form cells. Western blot analysis also indicated that p74...

Reproduction and Trypanosoma congolense in Nigerian West African Dwarf ewes:I. Effects on the estrous cycle OAK
Abubakar, Y.U.; Oyedipe, E.O.; Eduvie, L.O.; Ogwu, D.O.; Adeyeye, A.A..
Ten cycling Nigerian West African Dwarf (WAD) ewes were used to determine the effect of Trypanosoma congolense infection on the estrous cycle. The estrus of ewes were synchronized using intravaginal progesterone sponges and divided into 2 groups (A and B) of 5 ewes each after 54 days (3 cycles). The ewes in group A were inoculated with blood containing approximately 5×105 trypanosomes per ml of blood while ewes in group B served as the uninfected control. All the ewes were monitored from infection till the end of the study (56 days). Ewes in the infected group had silent estrus with clinical trypanosomosis that was characterized by undulating parasitemia, intermittent pyrexia, anemia and weight loss. There was no statistical (p > 0.05) difference...
